(Reuters) -Novavax Inc said on Friday it had started working on a version of its COVID-19 vaccine to target the variant detected in South Africa and would have the shot ready for testing and manufacturing in the next few weeks.
The company's COVID-19 shot contains an actual version of the virus' spike protein that cannot cause disease but can trigger the immune system. The vaccine developer said it had started developing a spike protein specifically based on the known genetic sequence of the variant, B.1.1.529.
"The initial work will take a few weeks," a company spokesperson said. Shares of the company closed up nearly 9% on Friday.
Novavax's vaccine received its first emergency use approval earlier this month in Indonesia followed by the Philippines.
The company has said it is on track to file for U.S. approval by the end of the year. It has also filed for approvals with the European Medicines Agency as well as in Canada.
Other vaccine developers, including Germany's BioNTech SE and Johnson & Johnson, have said they are testing the effectiveness of their shots against the new variant, which is named Omicron by the World Health Organisation.
Inovio Pharmaceuticals Inc said it had begun testing its vaccine candidate, INO-4800, to evaluate its effectiveness against the new variant. The company expects the testing to take about two weeks.
Inovio also said it was simultaneously designing a new vaccine candidate that specifically targeted Omicron.

For a bit of context, recently, I auditioned as a writer for a particularly big play (musical) in my school. Three people passed, and I am one of those three people.
Before we started writing, we had to discuss how the plot of the story was going to go on. I had the idea and made the base. I know that inevitably, the plot was going to be workshopped again before we started writing.
So, us three writers and the directors started discussing about the story. During the discussion, they began having ideas and started to change and add additional plots that from my perspective were not the greatest. I have been studying stage play for quite some time and I am quite confident with this judgement of mine. Their ideas felt forced and too complicated to write (it didn't connect with the plot). However, it was five people against one and I was clearly unheard and was on the losing side, so I chose to not complain and just going along with whatever they're saying.
Even after the discussion was finished, I still couldn't comprehend what their ideas were. So I just told them that "I won't write these certain parts that I don't understand." I basically would 'take' myself out of the narratives and let the other two writers write it, but they told me that I didn't even try to understand the plot.
I've been trying to understand the plot of the story but it just wouldn't process in my brain. It just didn't make sense and truthfully, I absolutely despised the new script they came up with.
So, right now, I'm trying to look for ways on how to be more open-minded and accepting of the story. Any suggestions?

Alright, so we all know the reaction the market had to Compass Pathway's phase IIb results. There's some discussion going around that this was in large part due to misinformed investors and that the results of the trial were actually quite positive. Perhaps people were expecting to be blown away like they might have been with the MAPS studies...perhaps it was the adverse events... perhaps psychedelics have simply been over-marketed as miracle drugs and the reality did not meet expectations. Or maybe the results truly ARE bad. Regardless of the reasons, I think it would help to place the study in more context.
To start with, let's look at the indication Compass is targeting: treatment resistant depression (TRD).
Treatment resistant depression: a difficult condition with few available options
Treatment resistant depression is loosely defined as a person suffering from depression who has failed at least 2 prior treatments. According to one study, 30% of patients with TRD will attempt suicide at least once in their lifetime, and compared to "regular" depression, they suffer 2x the hospitalization rate and more comorbidity like heart failure, diabetes, etc.
TRD is considered an "unmet medical need", which means there is no treatment that is adequately addressing it. It is also a global health problem with approx. 100 million people worldwide suffering from it.
Currently, there are only two drugs approved specifically to treat TRD: Symbax and J&J's SPRAVATO. SPRAVATO is the nasal spray version of esketamine, and, not only is it one of two options, it is the only "psychedelic" currently on the market that is approved by the FDA.
To illustrate how difficult the TRD indication is to treat, I urge you to read up on the SPRAVATO trials. Two out of four of their phase 3 trials failed to meet their primary endpoint goals! Yet it was still approved by the FDA. This goes to show how desperately new drugs are needed for this population.
More context: psilocybin study for TRD (2018) by Robin Carhart-Harris
While it was a small study of 20 patients, "Psilocybin with psychological support for treatment-resistant depression: six-month follow-up". is useful to look at. In this study, 9 patients (45%) were considered "responders" to psilocybin and 4 (20%) went into remission at week 5. Two doses of psilocybin were used, within a one week span, at 10mg and 25mg. Now the Compass' study had 233 patients. One single dose was used, either 10mg or 25mg. At week 6, 33% were "responders" and 25% went into remission in the 25mg group.
Adverse events: nothing new here
One of the biggest concerns surrounding the Compass study were the adverse events. Again, without any context, they may seem alarming. But we already have a useful study to compare & reference: another Robin Carhart-Harris' study called "Trial of Psilocybin versus Escitalopram for Depression". This was the last landmark study on psilocybin and was published in the New England Journal of Medicine. The adverse events were recorded in the supplementary appendix of this study and we can compare them to the Compass study.
Below are the adverse events found in >15% of Compass' treatment group (CMPS), v. the Robin Carhart-Harris study (RCH):

  1. Headache: 34.2% (CMPS) v. 66.7% (RCH)
  2. Nausea: 22.8% (CMPS) v. 26.7% (RCH)
  3. Fatigue 15.2% (CMPS) v. 6.7% (RCH)
Now let's also look at SPRAVATO trials. Remember, this is one of only two drugs available for TRD. Yet the # of side effects found in >15% of those treated were double:
  1. Nausea: 28.3%
  2. Dizziness: 23.7%
  3. Vertigo: 22.5%
  4. Headache: 20.2%
  5. Dysgeusia (altered taste): 18.8%
  6. Somnolence (drowsiness): 17.3%
Suicidal ideation & behavior
IMO this was the biggest concern of the trial. Any number of patients reporting suicidal thoughts & behavior would scare investors. But remember, this population is especially vulnerable to suicide. In fact, three patients were lost to suicide in the SPRAVATO trials. In addition, both the SPRAVATO and Compass trials showed rates of suicidal ideation as TESAEs (treatment emergent serious adverse events):
  1. SPRAVATO: 10.2% in treatment group; 12.3% in placebo group
  2. CMPS: 2.5% in 25mg treatment group; 0% placebo group
Suicidal behavior is the other issue and probably the most concerning of all. While we don't have all the data yet, we know that there were 3 suicidal behavior TESAEs in the Compass' 25mg group. We also know that these events happened at least one month after the drug administration. There is some speculation that these patients felt despair when the drug did not work for them. We can only wait to find out as more data is released and how Compass will address it in future studies.
33% response rate and 25% remission rate from one single dose
This is my last point. Remember that the 33% response rate and 25% remission rate were held at 3 months out (12 weeks) all from one single dose. This is a clear advantage over traditional, big pharma type chronic use drugs. This is not something you have to take every day, or even every month. This is not something you have to wait 4-6 weeks to get the benefit from, like an SSRI. Look again at SPRAVATO, where the dosing regimen is 2x a week for 4 weeks, and then maintenance dosing once every 1-2 weeks, all done under clinical supervision. SPRAVATO users have to experience the same adverse side effects, week after week after week, while having to schedule clinic visits, week after week after week. The benefits of psilocybin (or any other psychedelic) begin to clearly outweigh any current TRD treatment, especially from a long view perspective.
